GemPharmatech offers the urate oxidase knockout (Uox-KO) model for the study of gout and therapeutics targeting hyperuricemia.
Study Outline
Figure 1. Study Outline of Gout Models.
Validation Data
1. Uric acid level of Uox-KO mice
Figure 2. Both male and female mice spontaneously developed hyperuricemia. Females showed lower mean uric acid levels and better individual homogeneity than males. In Uox knockout mice, allopurinol effectively reduced hyperuricemia.
2. Renal function indices of Uox-KO mice
Figure 3. The elevated creatinine and urea nitrogen levels in Uox-KO male and female mice indicated the development of renal injury, which was more severe in males than females. Allopurinol (G4 group) did not alleviate the renal impairment induced by Uox knockout.
3. HE & Masson staining
Figure 4. Histopathology of the kidneys of Uox-KO mice revealed interstitial fibrous tissue hyperplasia with inflammatory cell infiltration, glomerular atrophy with thylakoid tissue hyperplasia, thickened vessel walls, dilated renal tubules forming cysts, and homogeneous red-stained substance exuding from the tubular lumen of the kidneys. Allopurinol (G4 group) did not improve the renal lesions caused by Uox knockout in mice.
