3. 항체 결합 분석

인간화 세포주 및 인간화 마우스 내에 표적이 존재하는 항체 기반 약물의 결합 효율을 평가하는 데 활용되는 이 방법은 시험 물질의 in vivo 효과에 대한 기준이 되는 양성 대조군과 시험 물질 간에 효과를 비교합니다.



3.1 A431 세포에서 세툭시맙(cetuximab) 결합 분석

세툭시맙 농도가 낮을 때에는 약물 농도가 증가하면서 세포에 결합된 약물의 양도 증가하여 검출되는 형광 신호가 증가하였습니다. 그러나 약물 농도가 포화 농도 이후에도 증가하면 검출되는 형광 신호는 증가하지 않았습니다.


3.2 MC38-hPDL1 세포에서 티쎈트릭(tecentriq) 결합 분석

티쎈트릭은 hPDL1을 표적화하는 항체 약물입니다. 티쎈트릭을 MC38-hPDL1 세포(젬파마텍이 변형한 세포주)와 함께 배양한 후 형광을 내는 2차 항체인 Anti-human IgG 항체를 가하여 유세포분석기로 약물과 세포 간의 결합 효율을 조사하였습니다.
